No, Galaxy Z TriFold does not support S Pen.

This is possible with a feature called App Continuity and it can be enabled through Settings.• The App Continuity feature allows an easy transition of your view between the cover screen and the larger main screen.• Enabling this feature will adjust the cover screen resolution slightly, but upscaling technology is applied to ensure the same or similar image quality as before.• However, when taking a screenshot on the cover screen, upscaling is not supported so the image resolution for your screenshots will be reduced.
